James Hamilton, 3. Earl of Arran (* 1537; † 1609) war ein schottischer Adliger.
James Hamilton, Sohn des James Hamilton, 2. Earl of Arran, warf sich in Frankreich den Hugenotten in die Arme, weshalb ihm der König von Frankreich sein Herzogtum Châtellerault entzog. Hamilton lebte hierauf als Prediger in Schottland. Er war als möglicher Ehemann für die schottische Königin Maria Stuart, als auch die englische Königin Elisabeth im Gespräch, verfiel aber 1561 dem Wahnsinn.
1575 erbte Hamilton nach dem Tode seines Vaters dessen Titel. Wegen seiner Geisteskrankheit wurde ihm jedoch sein Bruder John als Vormund zur Seite gestellt. James Hamilton starb 1609.
Nachdem Morton 1581 unter Jakob VI., dem nachmaligen König Jakob I. von England, das Schafott besteigen musste, wurden die Güter der Familie Hamilton eingezogen, sie selbst wurden geächtet, der Adelstitel aberkannt. Das Stammschloß wurde 1579 zerstört. Erst 1585 erhielt er seinen Titel zurück.
Der Bruder des 3. Earl of Arran, Claude wurde der Stifter der Seitenlinie der Marquess of Abercorn.
