Der Tuapeka River ist ein Fluss in der Region Otago auf der der Südinsel Neuseelands. Er ist ein Nebenfluss des Clutha River, in den er bei Tuapeka Mouth zwischen Roxburgh und Balclutha mündet.

Der Fluss ist hauptsächlich wegen seiner Rolle als Zentrum des Otago-Goldrausches der 1860er Jahre bekannt. Der erste bedeutende Goldfund in Otago war bei Gabriel's Gully (45° 52′ 54″ S, 169° 40′ 47″ O-45.881763888889169.67963333333) nahe Lawrence im Jahre 1861. Der Ort Tuapeka Mouth liegt südwestlich der Mündung des Flusses in den Clutha.
