Die Insulin-like growth factors (IGF) (deutsch Insulinähnliche Wachstumsfaktoren) sind Polypeptide, die eine hohe Sequenzhomologie zu Insulin zeigen. Sie sind Teil eines komplexen Systems, das Körperzellen nutzen, um mit ihrer Umgebung zu kommunizieren. Dieses System, das oft auch als IGF-Achse bezeichnet wird, besteht aus zwei Membranrezeptoren (IGF1R und IGF2R), zwei Liganden, einer Gruppe von sechs IGF-Bindungsproteinen (IGFBP 1-6), sowie einigen IGFBP-assoziierten Proteasen.
Funktionen
Insulin-like growth factor 1 (IGF-1), auch Somatomedin C genannt, ist ein Wachstumsfaktor, der strukturell dem Insulin sehr ähnlich ist. Er wird hauptsächlich von der Leber nach Stimulation mit Wachstumshormon (Growth Hormone (GH), Somatotropin) sezerniert. In Studien konnte ein Einfluss von IGF-1 auf die Regulation sowohl von physiologischen Zuständen als auch von pathologischen Zuständen vor allem in der Krebsentwicklung nachgewiesen werden. Hierbei wurde ein Einfluss auf die Zellproliferation und die Apoptoseverhinderung beschrieben.
Insulin-like growth factor 2 (IGF-2), auch Somatomedin A genannt, scheint für die frühe Zellentwicklung eine wichtige Rolle zu spielen, also eventuell in der Fetalphase, während IGF-1 eher zu einem späteren Zeitpunkt für die Wachstumsmaximierung zuständig ist.
