Die Carboxylgruppe

Die Carboxylgruppe (auch Carboxygruppe) ist die funktionelle Gruppe der Carbonsäuren und hat die Formel -COOH.

Das Carboxylkohlenstoffatom trägt ein doppeltgebundenes Sauerstoff-Atom und eine einfach gebundene Hydroxylgruppe. Aufgrund der Elektronegativitätdifferenz zwischen dem Sauerstoff und Kohlenstoff trägt das Kohlenstoffatom eine positive Partialladung. Daher ist das Carboxylkohlenstoffatom leicht nukleophil angreifbar. Die Hydroxyleinheit der Carboxyl ist verhältnismäßig sauer, das Wasserstoffatom wird leicht an einen entsprechenden Partner abgegeben. Diese im Vergleich zu Alkoholen erhöhte Acidität (Säurestärke) liegt in der Resonanzstabilisierung der korrespondierenden Base, dem negativ geladenen Carboxylation begründet. Im Carboxylation sind die Sauerstoffatome gleichwertig, das heißt die negative Ladung ist auf beide Sauerstoffatome verteilt und beide C-O-Bindungen besitzen partiellen Doppelbindungscharakter.

Carbonsäuren liegen in Lösung und Kristall oft als Dimere vor, dabei werden Carbonyl- und Hydroxylsauerstoff zweier verschiedener Moleküle durch das Hydroxylproton wasserstoffverbrückt. Die Paarbildung erlaubt dabei die Ausbildung von zwei Wasserstoffbrücken pro Dimer.

Carboxylgruppen sind als Teil der Aminosäuren eine in der Natur häufigsten funktionellen Gruppen, und sind Vorläufer der Peptidbindung, mit der Aminosäuren zu Proteinen verbunden werden.
