James Johnstone, 2. Marquess of Annandale (* um 1687/1688; † 1730 in Neapel) 3. Earl of Annandale und Hartfell, sowie 2. Marquess of Annandale, wurde um 1687/8 geboren und war der älteste Sohn von William Johnstone, 2. Earl of Annandale und Hartfell, sowie 1. Marquess of Annandale, und seiner ersten Ehefrau Sophia Fairholm. Er erbte 1721 die Titel von seinem Vater.
Er verbrachte die meiste Zeit seines Lebens in Italien, wo er ein große Kunstsammlung und Antiquitäten ansammelte. 1726 versuchte er gesetzlich seinen Halbbruder daran zu hindern seine Peerage und den Grundbesitz zu erben. Diese sollten auf seine Schwester Henrietta, Countess of Hopetoun und ihre Nachkommen gehen. Jedoch war dieser Versuch nur zum Teil erfolgreich, da er 1730 in Italien, im Alter von 42 Jahren, verstarb. Nur seine schottischen Ländereien und seine Kunstsammlung gingen an Henrietta, während seine Peerage (welche nicht legitim übertragen werden konnte, so wie er es beabsichtigte) an seinen Halbbruder George überging, so dass er der 4. Earl of Annandale und Hartfell, sowie 3. Marquess of Annandale wurde. Sein Leichnam wurde aus Italien überführt und in der Westminster Abbey beigesetzt. Sein Porträt hängt heute im Hopetoun House nahe Edinburgh.
