Hygromycin B ist ein Antibiotikum welches von Bakterien der Art Streptomyces hygroscopicus produziert wird. Es handelt sich dabei um ein Aminoglycosid, welches für Bakterien, Pilze und Zellen höherer Eukaryoten toxisch ist. Die Wirkung beruht auf einer Inhibierung der Proteinbiosynthese.[3]

Geschichte

Ursprünglich wurde Hygromycin B in den 1950er-Jahren entwickelt. Es findet seitdem Anwendung als Nahrungszusatz bei der Zucht von Schweinen und Hühnern als Antiwurm-Agenz (Handelsname: Hygromix). Die Hygromycin B produzierende Bakterien Art Streptomyces hygroscopicus wurde 1953 aus einer Bodenprobe isoliert. Resistenzgene wurden in den frühen 80er-Jahren des vorigen Jahrhunderts entdeckt.[4][5]

Verwendung in der Forschung

Im Laboralltag findet das Antibiotikum Anwendung bei der Selektion prokaryotischer und eukaryotischer Zellen mit Hygromycin-Resistenz-Gen. Die Resistenz wird von einer Kinase vermittelt welche Hygromycin B durch Phosphorylierung inaktiviert.[6] Seit der Entdeckung des Hygromycin-Resistenz-Gens wird Hygromycin B als eines der Standard-Antibiotika zur Selektion verwendet.
