Vorinostat ist eine aromatische chemische Verbindung aus der Gruppe der Oxime, als oral zu applizierender Arzneistoff zur Histon-Desacetylase-Inhibition eingesetzt wird. Es ist für die Behandlung des cutanen T-Zelllymphoms zugelassen. Weitere aktuelle klinische Studien betreffen die Wirkung von Vorinostat bei Darmkrebs und anderen Krebsarten sowie in Kombinationstherapien bei soliden Tumoren.

Pharmakologie

Histone stellen den Proteinanteil des Chromatins dar. Mittels Acetylierung und Desacetylierung der Histone wird die Transkription der DNA reguliert. Eine Hemmung der Histon-Desacetylase hat eine Hyperacetylierung der Histone zur Folge. Dies kann u.a. zu einem kontrollierten Zelltod (Apoptose) der Krebszellen führen.
