Bei der Volkszählung im Jahr 2000 hatte der County 470.658 Einwohner und eine Bevölkerungsdichte von 191 Einwohner pro Quadratkilometer. Der Verwaltungssitz (County Seat) ist Lancaster. Besonders bekannt ist Lancaster County als Siedlungsort der Glaubensgemeinschaft der deutschstämmigen Amischen. Etwa fünf Prozent der Bevölkerung sprechen als Umgangssprache Pennsylvania Dutch.

Geschichte
Das County wurde am 10. Mai 1729 aus Chester County gebildet und nach Lancashire, England benannt.

Das County hat eine Fläche von 2.548 Quadratkilometern, wovon 90 Quadratkilometer Wasserfläche sind.
